Retatrutide is a synthetic 39-residue peptide that adds glucagon receptor activity to the dual incretin design, making it a triple agonist. The backbone carries Aib and α-methyl-leucine substitutions for protease resistance, plus a C20 diacid-γGlu-AEEA linker at position 17 for albumin binding.

It is used in laboratories to compare potency and efficacy across three receptor systems in the same assay plate, to study receptor-selectivity ratios, and in in-vitro models of energy substrate handling and glycemic homeostasis research. Side-by-side runs against a dual agonist such as tirzepatide are the usual way to isolate the glucagon-receptor contribution.
