Research Information
Tirzepatide is a synthetic 39-residue incretin analogue that engages both the GIP and the GLP-1 receptor. Two aminoisobutyric acid substitutions protect the N-terminus from DPP-4 cleavage, and a C20 fatty diacid chain conjugated at Lys20 drives albumin binding — the pair of modifications is what gives the molecule its long circulating profile in published pharmacokinetic work.
Laboratories use it as a reference agonist in receptor-binding and displacement assays, cAMP accumulation and β-arrestin recruitment readouts, comparative studies of biased signalling between the two receptors, and in-vitro models of glycemic homeostasis research. Because it activates two receptors with different potency ratios, it is also a common control when characterising newer multi-agonists.
Supplied as a lyophilised powder under vacuum. Reconstitute with bacteriostatic water, swirl rather than shake, and keep the reconstituted solution refrigerated and protected from light. Aliquot before freezing to avoid repeated freeze-thaw cycles.
